Principle of the assay: TFPI ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-TFPI antibody and an TFPI-HRP conjugate. The assay sample and buffer are incubated together with TFPI-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the TFPI concentration since TFPI from samples and TFPI-HRP conjugate compete for the anti-TFPI antibody binding site. Since the number of sites is limited, as more sites are occupied by TFPI from the sample, fewer sites are left to bind TFPI-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The TFPI concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a protease inhibitor that regulates the tissue factor (TF)-dependent pathway of blood coagulation. The coagulation process initiates with the formation of a factor VIIa-TF complex, which proteolytically activates additional proteases (factors IX and X) and ultimately leads to the formation of a fibrin clot. The product of this gene inhibits the activated factor X and VIIa-TF proteases in an autoregulatory loop. The encoded protein is glycosylated and predominantly found in the vascular endothelium and plasma in both free forms and complexed with plasma lipoproteins. Several alternatively spliced transcript variants of this gene have been described, but the full-length nature of some of these variants has not been confirmed. [provided by RefSeq, Jul 2008]

Uniprot Description

TFPI: Inhibits factor X (X(a)) directly and, in a Xa-dependent way, inhibits VIIa/tissue factor activity, presumably by forming a quaternary Xa/LACI/VIIa/TF complex. It possesses an antithrombotic action and also the ability to associate with lipoproteins in plasma. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes

The Goat TFPI tfpi (Catalog #AAA735288) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA735288 ELISA Kit recognizes Goat TFPI. It is sometimes possible for the material contained within the vial of "Tissue factor pathway inhibitor, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
