Principle of the Assay: TF ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-TF antibody and an TF-HRP conjugate. The assay sample and buffer are incubated together with TF-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the TF concentration since TF from samples and TF-HRP conjugate compete for the anti-TF antibody binding site. Since the number of sites is limited, as more sites are occupied by TF from the sample, fewer sites are left to bind TF-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The TF concentration in each sample is interpolated from this standard curve.

This gene encodes a glycoprotein with an approximate molecular weight of 76.5 kDa. It is thought to have been created as a result of an ancient gene duplication event that led to generation of homologous C and N-terminal domains each of which binds one ion of ferric iron. The function of this protein is to transport iron from the intestine, reticuloendothelial system, and liver parenchymal cells to all proliferating cells in the body. This protein may also have a physiologic role as granulocyte/pollen-binding protein (GPBP) involved in the removal of certain organic matter and allergens from serum. [provided by RefSeq, Sep 2009]

It is responsible for the transport of iron from sites of absorption and heme degradation to those of storage and utilization. Serum transferrin may also have a further role in stimulating cell proliferation. Defects in TF are the cause of atransferrinemia (ATRAF). Atransferrinemia is rare autosomal recessive disorder characterized by iron overload and hypochromic anemia. Belongs to the transferrin family.
