This assay has high sensitivity and excellent specificity for detection of TDO2,3. No significant cross-reactivity or interference between TDO2,3 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between TDO2,3 and all the analogues, therefore, cross reaction may still exist in some cases.

Intended Uses: This TDO2,3 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of MouseTDO2,3. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: TDO2,3 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-TDO2,3 antibody and an TDO2,3-HRP conjugate. The assay sample and buffer are incubated together with TDO2,3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the TDO2,3 concentration since TDO2,3 from samples and TDO2,3-HRP conjugate compete for the anti-TDO2,3 antibody binding site. Since the number of sites is limited, as more sites are occupied by TDO2,3 from the sample, fewer sites are left to bind TDO2,3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The TDO2,3 concentration in each sample is interpolated from this standard curve.

This gene encodes a heme enzyme that plays a critical role in tryptophan metabolism by catalyzing the first and rate-limiting step of the kynurenine pathway. Increased activity of the encoded protein and subsequent kynurenine production may also play a role in cancer through the suppression of antitumor immune responses, and single nucleotide polymorphisms in this gene may be associated with autism.
