Intended Uses: This TCN2 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Canine TCN2. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: TCN2 ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-TCN2 antibody and an TCN2-HRP conjugate. The assay sample and buffer are incubated together with TCN2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the TCN2 concentration since TCN2 from samples and TCN2-HRP conjugate compete for the anti-TCN2 antibody binding site. Since the number of sites is limited, as more sites are occupied by TCN2 from the sample, fewer sites are left to bind TCN2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The TCN2 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a member of the vitamin B12-binding protein family. This family of proteins, alternatively referred to as R binders, is expressed in various tissues and secretions. This plasma protein binds cobalamin and mediates the transport of cobalamin into cells. This protein and other mammalian cobalamin-binding proteins, such as transcobalamin I and gastric intrisic factor, may have evolved by duplication of a common ancestral gene. Alternative splicing results in multiple transcript variants.[provided by RefSeq, May 2010]
