Quality Control
Oligonucleotide synthesis is monitored base by base through trityl analysis to ensure appropriate coupling efficiency. The oligo is subsequently purified by affinity-solid phase extraction. The annealed RNA duplex is further analyzed by mass spectrometry to verify the exact composition of the duplex. Each lot is compared to the previous lot by mass spectrometry to ensure maximum lot-to-lot consistency.
Directions for Use
We recommends transfection with 100 nM siRNA 48 to 72 hours prior to cell lysis. Before resuspending, briefly centrifuge the tube to ensure the lyophilized siRNA is at the bottom of the tube. Resuspend the siRNA oligos to an appropriate concentration with DEPC water. For each vial, suitable for 250 transfections in 24 well plate (20 pmol for each well).
Components
We offer pre-designed sets of 3 different target-specific siRNA oligo duplexes of rat GUCY2C gene. Each vial contains 5 nmol of lyophilized siRNA. The duplexes can be transfected individually or pooled together to achieve knockdown of the target gene, which is most commonly assessed by qPCR or western blot. Our siRNA oligos are also chemically modified (2'-OMe) at no extra charge for increased stability and enhanced knockdown in vitro and in vivo.
Preparation and Storage
Shipped at 4 degree C. Store at -20 degree C for one year.

NCBI Description

guanylyl cyclase; functions as receptor for heat-stable enterotoxin responsible for acute diarrhea [RGD, Feb 2006]

Uniprot Description

GUCY2C: Receptor for the E.coli heat-stable enterotoxin (E.coli enterotoxin markedly stimulates the accumulation of cGMP in mammalian cells expressing GC-C). Also activated by the endogenous peptide guanylin. Defects in GUCY2C are the cause of diarrhea type 6 (DIAR6). A relatively mild, early-onset chronic diarrhea that may be associated with increased susceptibility to inflammatory bowel disease, small-bowel obstruction, and esophagitis. Defects in GUCY2C are a cause of meconium ileus (MECIL). A condition characterized by a intestinal obstruction due to inspissated meconium in the distal ileum and cecum, which develops in utero and presents shortly after birth as a failure to pass meconium. Meconium ileus is a known clinical manifestation of cystic fibrosis. Belongs to the adenylyl cyclase class-4/guanylyl cyclase family.

Protein type: Protein kinase, dual-specificity (receptor); Guanylyl cyclase; Nucleotide Metabolism - purine; Receptor, misc.; Kinase, protein; Protein kinase, RGC; EC 4.6.1.2; Lyase; Membrane protein, integral; RGC group; RGC family

Cellular Component: endoplasmic reticulum membrane; integral to membrane; plasma membrane

Molecular Function: toxin binding; guanylate cyclase activity; GTP binding; ATP binding; protein kinase activity

Biological Process: cGMP biosynthetic process; response to toxin; protein amino acid phosphorylation; regulation of cell proliferation

Product Notes

The GUCY2C gucy2c (Catalog #AAA8202050) is a siRNA produced from Synthetic and is intended for research purposes only. The product is available for immediate purchase. The GUCY2C siRNA (Rat) reacts with Rat and may cross-react with other species as described in the data sheet. AAA Biotech's GUCY2C can be used in a range of immunoassay formats including, but not limited to, RNA Interference (RNAi). Researchers should empirically determine the suitability of the GUCY2C gucy2c for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "GUCY2C, siRNA" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
