RNA Interference (RNAi)
Quality Control
Oligonucleotide synthesis is monitored base by base through trityl analysis to ensure appropriate coupling efficiency. The oligo is subsequently purified by affinity-solid phase extraction. The annealed RNA duplex is further analyzed by mass spectrometry to verify the exact composition of the duplex. Each lot is compared to the previous lot by mass spectrometry to ensure maximum lot-to-lot consistency.
Directions for Use
We recommends transfection with 100 nM siRNA 48 to 72 hours prior to cell lysis. Before resuspending, briefly centrifuge the tube to ensure the lyophilized siRNA is at the bottom of the tube. Resuspend the siRNA oligos to an appropriate concentration with DEPC water. For each vial, suitable for 250 transfections in 24 well plate (20 pmol for each well).
Components
We offer pre-designed sets of 3 different target-specific siRNA oligo duplexes of mouse TBL1X gene. Each vial contains 5 nmol of lyophilized siRNA. The duplexes can be transfected individually or pooled together to achieve knockdown of the target gene, which is most commonly assessed by qPCR or western blot. Our siRNA oligos are also chemically modified (2'-OMe) at no extra charge for increased stability and enhanced knockdown in vitro and in vivo.
Preparation and Storage
Shipped at 4 degree C. Store at -20 degree C for one year.

Uniprot Description

TBL1X: F-box-like protein involved in the recruitment of the ubiquitin/19S proteasome complex to nuclear receptor-regulated transcription units. Plays an essential role in transcription activation mediated by nuclear receptors. Probably acts as integral component of corepressor complexes that mediates the recruitment of the 19S proteasome complex, leading to the subsequent proteasomal degradation of transcription repressor complexes, thereby allowing cofactor exchange. Belongs to the WD repeat EBI family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Transcription, coactivator/corepressor; Nuclear receptor co-regulator

Cellular Component: transcription factor complex; transcriptional repressor complex; spindle microtubule; histone deacetylase complex; nucleus

Molecular Function: protein C-terminus binding; protein domain specific binding; protein binding; DNA binding; histone binding; beta-catenin binding; chromatin binding; transcription factor binding; transcription corepressor activity

Biological Process: fat cell differentiation; proteasomal ubiquitin-dependent protein catabolic process; transcription, DNA-dependent; positive regulation of transcription, DNA-dependent; histone deacetylation; Wnt receptor signaling pathway through beta-catenin; negative regulation of transcription from RNA polymerase II promoter; proteolysis; regulation of transcription from RNA polymerase II promoter; sensory perception of sound; regulation of transcription, DNA-dependent; response to steroid hormone stimulus; response to estrogen stimulus; positive regulation of transcription from RNA polymerase II promoter; negative regulation of transcription, DNA-dependent

Product Notes

The TBL1X tbl1x (Catalog #AAA829996) is a siRNA produced from Synthetic and is intended for research purposes only. The product is available for immediate purchase. The TBL1X siRNA (Mouse) reacts with Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's TBL1X can be used in a range of immunoassay formats including, but not limited to, RNA Interference (RNAi). Researchers should empirically determine the suitability of the TBL1X tbl1x for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "TBL1X, siRNA" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
