STXBP3: a vesicle protein that, together with STX4 and VAMP2, regulates the insulin-dependent movement of GLUT4 and the docking/fusion of intracellular GLUT4-containing vesicles with the cell surface in adipocytes. Reduction in its expression using siRNA impairs insulin-stimulated GLUT4 vesicle exocytosis in 3T3-L1 adipocytes. Phosphorylated by the insulin receptor (InsR) at Tyr521 in vitro, and bound and phosphorylated by the activated InsR within adipocytes and skeletal muscle. Tyrosine phosphorylation on residues Y219 and Y521 correlates with its dissociation from STX4. Phosphorylated by PKC in platelets in response to thrombin stimulation, inhibiting its binding to STX4. Interacts directly with DOC2B at the cell membrane, excluding its interaction with STX4, regulating glucose-stimulated insulin secretion. Belongs to the STXBP/unc-18/SEC1 family.
