Recombinant full-length human STK16 was expressed by baculovirus in Sf9 insect cells using an N-terminal GST tag.

Scientific Background: STK16 is serine/threonine kinase 16 which constitute the catalytic subunit of a multisubunit kinase. STK16 is ubiquitously expressed at very low levels and expression of an STK16 fusion protein showed that STK16 is localized with Golgi and Golgi-derived vesicles. The overexpression of STK16 leads to disorganization of the Golgi apparatus into vesicular structures and STK16 is highlyinvolved in the secretory pathway (1). STK16 is involved in the transforming growth factor (TGF)-beta signalling pathway (2).

Uniprot Description

Function: Membrane-associated protein kinase that phosphorylates on serine and threonine residues. In vitro substrates include DRG1, ENO1 and EIF4EBP1. Also autophosphorylates. May be involved in secretory vesicle trafficking or intracellular signaling. May have a role in regulating stromal-epithelial interactions that occur during ductal morphogenesis in the mammary gland. May be involved in TGF-beta signaling. Able to autophosphorylate on Tyr residue; it is however unclear whether it has tyrosine-protein kinase toward other proteins. Ref.2

Catalytic activity: ATP + a protein = ADP + a phosphoprotein.ATP + a [protein]-L-tyrosine = ADP + a [protein]-L-tyrosine phosphate.

Subunit structure: Monomer. Interacts with DRG1 (via its N-terminal); the interaction phosphorylates DRG1. Ref.10
