Intended Uses: This SSTR1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Porcine SSTR1. This ELISA kit for research use only!

Principle of the Assay: SSTR1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-SSTR1 antibody and an SSTR1-HRP conjugate. The assay sample and buffer are incubated together with SSTR1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the SSTR1 concentration since SSTR1 from samples and SSTR1-HRP conjugate compete for the anti-SSTR1 antibody binding site. Since the number of sites is limited, as more sites are occupied by SSTR1 from the sample, fewer sites are left to bind SSTR1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The SSTR1 concentration in each sample is interpolated from this standard curve.

NCBI Description

Somatostatins are peptide hormones that regulate diverse cellular functions such as neurotransmission, cell proliferation, and endocrine signaling as well as inhibiting the release of many hormones and other secretory proteins. Somatostatin has two active forms of 14 and 28 amino acids. The biological effects of somatostatins are mediated by a family of G-protein coupled somatostatin receptors that are expressed in a tissue-specific manner. The protein encoded by this gene is a member of the superfamily of somatostatin receptors having seven transmembrane segments. Somatostatin receptors form homodimers and heterodimers with other members of the superfamily as well as with other G-protein coupled receptors and receptor tyrosine kinases. This somatostatin receptor has greater affinity for somatostatin-14 than -28. [provided by RefSeq, Jul 2012]
