Principle of the assay: Smad4 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-Smad4 antibody and an Smad4-HRP conjugate. The assay sample and buffer are incubated together with Smad4-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the Smad4 concentration since Smad4 from samples and Smad4-HRP conjugate compete for the anti-Smad4 antibody binding site. Since the number of sites is limited, as more sites are occupied by Smad4 from the sample, fewer sites are left to bind Smad4-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The Smad4 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a member of the Smad family of signal transduction proteins. Smad proteins are phosphorylated and activated by transmembrane serine-threonine receptor kinases in response to TGF-beta signaling. The product of this gene forms homomeric complexes and heteromeric complexes with other activated Smad proteins, which then accumulate in the nucleus and regulate the transcription of target genes. This protein binds to DNA and recognizes an 8-bp palindromic sequence (GTCTAGAC) called the Smad-binding element (SBE). The Smad proteins are subject to complex regulation by post-translational modifications. Mutations or deletions in this gene have been shown to result in pancreatic cancer, juvenile polyposis syndrome, and hereditary hemorrhagic telangiectasia syndrome. [provided by RefSeq, Oct 2009]
