Preparation and Storage

Principle of the Assay||This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-Human SLAM/CD150 antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-Human SLAM/CD150 antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. HRP-Streptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Human SLAM/CD150 amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of Human SLAM/CD150 can be calculated.

Uniprot Description

SLAM: a type I membrane protein present on the surface of B cells, T cells and CD83+ dendritic cells. A high-affinity self-ligand considered to be important in bidirectional T <-> B-cell stimulation. SLAM-induced signal-transduction events in T lymphocytes are different from those in B cells. Two modes of SLAM signaling are likely to exist: one in which the inhibitor SH2D1A (SAP) acts as a negative regulator and another in which protein-tyrosine phosphatase 2C (SHP-2)-dependent signal transduction operates. Its cytoplasmic domain interacts with SAP through part of its SH2 domain, and with SHP-2. Three alternatively spliced isoforms have been described.
