Related Product Information for SERPINE1 elisa kit
Principle of the Assay: The Picokine™ Human SERPINE1 Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid-phase immunoassay specially designed to measure Human SERPINE1 with a 96-well strip plate that is pre-coated with antibody specific for SERPINE1. The detection antibody is a biotinylated antibody specific for SERPINE1. The capture antibody is monoclonal antibody from mouse and the detection antibody is polyclonal antibody from goat. The kit contains recombinant Human SERPINE1 with immunogen: Expression system for standard: sf21; Immunogen sequence: S22-P402. The kit is analytically validated with ready-to-use reagents. To measure Human SERPINE1, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is an HRP substrate and will be catalyzed to produce a blue color product, which changes into yellow after adding the acidic stop solution. The absorbance of the yellow product at 450nm is linearly proportional to Human SERPINE1 in the sample. Read the absorbance of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Human SERPINE1 in the sample.

Background/Introduction: Plasminogen activator inhibitor-1 is the principal inhibitor of tissue plasminogen activator (tPA) and urokinase (uPA), the activators of plasminogen and hence fibrinolysis (the physiological breakdown of blood clots). It is a serine protease inhibitor (serpin) protein (SERPINE1). PAI-1 is mainly produced by the endothelium (cells lining blood vessels), but is also secreted by other tissue types, such as adipose tissue. The PAI-1 gene is located on the seventh chromosome (7q21.3-q22).1 Congenital deficiency of PAI-1 leads to a hemorrhagic diathesis (a tendency to hemorrhage). PAI-1 is present in increased levels in various disease states (such as a number of forms of cancer), as well as in obesity and the metabolic syndrome. It has been linked to the increased occurrence of thrombosis in patients with these conditions. Additionally, it has been shown to interact with ORM1.2 The standard product used in this kit is recombinant human PAI-1 with the molecular mass of 43KDa.

NCBI Description

This gene encodes a member of the serine proteinase inhibitor (serpin) superfamily. This member is the principal inhibitor of tissue plasminogen activator (tPA) and urokinase (uPA), and hence is an inhibitor of fibrinolysis. Defects in this gene are the cause of plasminogen activator inhibitor-1 deficiency (PAI-1 deficiency), and high concentrations of the gene product are associated with thrombophilia. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Sep 2009]

Uniprot Description

SERPINE1: a secreted protein that acts as 'bait' for tissue plasminogen activator, urokinase, and protein C. Its rapid interaction with TPA may function as a major control point in the regulation of fibrinolysis. Belongs to the serpin family. Interacts with VTN. Binds LRP1B; binding is followed by internalization and degradation. Plasma levels of PAI-1 and VCAM-1 together may be useful in predicting post-operative recurrence in patients with colorectal cancer.

Product Notes

The Human SERPINE1 serpine1 (Catalog #AAA175945) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA175945 ELISA Kit recognizes Human SERPINE1. It is sometimes possible for the material contained within the vial of "PAI-1, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
