Principle of the Assay: Human Serpin A5/PAI3 ELISA Kit was based on standard sandwich enzyme-linked immune- sorbent assay technology. A monoclonal antibody from mouse specific for Serpin A5/PAI3 has been precoated onto 96-well plates. Standards(Expression system for standard: NSO, Immunogen sequence: E26-P406) and test samples are added to the wells, a biotinylated detection biotinylated polyclonal antibody from goat specific for Serpin A5/PAI3 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Human Serpin A5/PAI3 amount of sample captured in plate.

It is a 52kD glycoprotein and belongs to serine protease inhibitor (Serpin) super family of protein. This family member is a glycoprotein that can inhibit several serine proteases, including protein C and various plasminogen activators and kallikreins, and it thus plays diverse roles in hemostasis and thrombosis in multiple organs. This gene is mapped on the q arm of chromosome 14.

Heparin-dependent serine protease inhibitor acting in body fluids and secretions. Inactivates serine proteases by binding irreversibly to their serine activation site. Involved in the regulation of intravascular and extravascular proteolytic activities. Plays hemostatic roles in the blood plasma. Acts as a procoagulant and proinflammatory factor by inhibiting the anticoagulant activated protein C factor as well as the generation of activated protein C factor by the thrombin/thrombomodulin complex.
