Principle of the Assay||The Human CD62E ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human CD62E in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Human CD62E coated on a 96-well plate. Standards and samples are pipetted into the wells and CD62E present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human CD62E antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of CD62E bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background/Introduction: E-selectin, also called endothelial-leukocyte adhesion molecule-1 (ELAM-1), is a cell surface glycoprotein expressed by cytokine-activated endothelium that mediates the adhesion of blood neutrophils. An increased expression of E-selectin has been observed in the arterial endothelium interacting with lymphocytes and macrophages in human atherosclerotic lesions. E-selectin plays a critical role in mediating tissue-specific homing of T cells into skin, and of primitive hematopoietic progenitor cells (HPCs) into bone marrow (BM). A structurally and functionally related group of genes, lymph node homing receptor (LHR), granule membrane protein 140 (GMP-140), and endothelial leukocyte adhesion molecule 1 (ELAM-1) are shown to constitute a gene cluster on mouse and human chromosome 1.

NCBI Description

The protein encoded by this gene is found in cytokine-stimulated endothelial cells and is thought to be responsible for the accumulation of blood leukocytes at sites of inflammation by mediating the adhesion of cells to the vascular lining. It exhibits structural features such as the presence of lectin- and EGF-like domains followed by short consensus repeat (SCR) domains that contain 6 conserved cysteine residues. These proteins are part of the selectin family of cell adhesion molecules. Adhesion molecules participate in the interaction between leukocytes and the endothelium and appear to be involved in the pathogenesis of atherosclerosis. [provided by RefSeq, Jul 2008]

Uniprot Description

SELE: Cell-surface glycoprotein having a role in immunoadhesion. Mediates in the adhesion of blood neutrophils in cytokine-activated endothelium through interaction with PSGL1/SELPLG. May have a role in capillary morphogenesis. Belongs to the selectin/LECAM family.

Product Notes

The Human SELE sele (Catalog #AAA8244699) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA8244699 ELISA Kit recognizes Human SELE. AAA Biotech's CD62E can be used in a range of immunoassay formats including, but not limited to, Sandwich ELISA (EIA). Researchers should empirically determine the suitability of the SELE sele for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CD62E, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
