Principle of the Assay||This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-Human RNLSantibody was pre-coated onto 96-well plates. And the biotin conjugated anti-Human RNLS antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. HRPStreptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Human RNLS amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration ofHuman RNLScan be calculated.

Renalase is a flavin adenine dinucleotide-dependent amine oxidase that is secreted into the blood from the kidney (Xu et al., 2005 [PubMed 15841207]).[supplied by OMIM, Mar 2008]

RENALASE: Probable FAD-dependent amine oxidase secreted by the kidney, which circulates in blood and modulates cardiac function and systemic blood pressure. Degrades catecholamines such as dopamine, norepinephrine and epinephrine in vitro. Lowers blood pressure in vivo by decreasing cardiac contractility and heart rate and preventing a compensatory increase in peripheral vascular tone, suggesting a causal link to the increased plasma catecholamine and heightened cardiovascular risk. High concentrations of catecholamines activate plasma renalase and promotes its secretion and synthesis. According to PubMed:17385068, is unlikely that renalase has physiologically relevant catecholamine-oxidizing activity. Belongs to the renalase family. 2 isoforms of the human protein are produced by alternative splicing.
