Principle of the Assay: RNF4 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-RNF4 antibody and an RNF4-HRP conjugate. The assay sample and buffer are incubated together with RNF4-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the RNF4 concentration since RNF4 from samples and RNF4-HRP conjugate compete for the anti-RNF4 antibody binding site. Since the number of sites is limited, as more sites are occupied by RNF4 from the sample, fewer sites are left to bind RNF4-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The RNF4 concentration in each sample is interpolated from this standard curve

NCBI Description

The protein encoded by this gene contains a RING finger motif and acts as a transcription regulator. This protein has been shown to interact with, and inhibit the activity of, TRPS1, a transcription suppressor of GATA-mediated transcription. Transcription repressor ZNF278/PATZ is found to interact with this protein, and thus reduce the enhancement of androgen receptor-dependent transcription mediated by this protein. Studies of the mouse and rat counterparts suggested a role of this protein in spermatogenesis. A pseudogene of this gene is found on chromosome 1.[provided by RefSeq, Jul 2010]
