Digestion of Agarose-embedded DNA
A minimum of 5units of enzyme is required for complete digestion of 1ug of agarose-embedded lambda DNA in 16 hours.

Overdigestion Assay
No detectable change in the specific fragmentation pattern is observed after 160-fold overdigestion (10u/ug lambda DNA x 16 hours) with BpiI.
Blue/White Cloning Assay
The B/W assay was relpaced with L0 test after validating experiments showed L0 test ability to detect nuclease and phosphatase activities with sensitivity that equals to that of B/W test.
Labeled Oligonucleotide (LO) Assay
No detectable degradation of single-stranded or double-stranded labeled oligonucleotide occurred duringincubation with 10 units of Bpil for 4 hours.
Ligation/Recutting Assay
The ligation and recleavage assay was replaced with L0 test after validating experiments showed L0 test ability to trace nuclease and phosphatase activities with sensitivity that is higher than L/R by a factor of 100.

Stability During Prolonged Incubation

Diluent Buffer
10mM Tris-HCl, pH 7.4 100mM KCl, 1mM EDTA, 1mM DTT, 0.2mg/ml BSA, 50% glycerol.
Thermal Inactivation
Bpil is inactivated by incubation at 65 degree C for 20 minutes.
Unit Definition
One unit is defined as the amount of BpiI required to digest 1ug of lambda DNA in 1 hour at 37 degree C in 50ul of reaction buffer.

MBS638383: Restriction Enzyme Buffer A, 10X, Dilute to 1X. 1X buffer compostion is 33mM Tris-acetate pH 7.9 at 37°C, 10mM magnesium acetate, 66mM potassium acetate, 0.1mg/ml BSA.
MBS638414: Restriction Enzyme Buffer C, 10X, Dilute to 1X for use. 1X buffer composition is 10mM Tris-HCl pH 7.5, 10mM MgCl2, 50mM sodium chloride, 0.1mg/ml BSA.
Protocol for Digestion
Water nuclease free: 16u

DNA(0.5-1ug/ul): 1 ul

Mix gently and spin down for a few seconds. Incubate at 37 degree C for 1-16 hours. The digestation reaction may be either scaled up or down.
Protocol for Digestion of PCR products Directly Amplification
PCR Reaction Mixture: 10ul (about 0.1-0.5ug of DNA)
Water, nuclease free: 18ul

Mix gently and spin down for a few seconds. Incubate at 37 degree C for 1-16 hours.
Preparation and Storage
May be stored at 4 degree C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Aliquots are stable for 6 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Basic phospholipase A2 BP-II

Uniprot Description

Snake venom phospholipase A2 (PLA2) that shows anticoagulant activities, strong myolytic activity, infiltration of polymorphonuclear cells, and edema in stromal tissues. Induces cell death of Jurkat cells in a concentration dependent manner. Shows a low phospholipase A2 activity. PLA2 catalyzes the calcium-dependent hydrolysis of the 2-acyl groups in 3-sn-phosphoglycerides.
