Procedural Notes
This purified product exhibits moderate enzymatic activity for Cumene hydroperoxide, the substrate that is most commonly used to assess the glutathione peroxidase activity of soluble GSTs. However, as is suspected for GST activity analysis using synthetic and natural substrates, low concentrations (high dilutions) of the enzyme may result in lower activity values. In contrast, initial velocities are much higher for more concentrated levels of enzyme but the rate decreases rapidly. Therefore, activity towards GST substrates and these considerations provide guidance when assaying this product under low concentrations or when using substrates other than Cumene hydroperoxide
Specific Activity
8.8 Units/mg as assayed by the spectrophotometric determination of NADPH oxidation coupled to the glutathione peroxidase activity of GST T1-1 on cumene hydroperoxide (1.5 mM) in the presence of reduced glutathione (1 mM) in 100 mM NaPO4 (pH 7.0) at room temperature.

NCBI Description

The protein encoded by this gene, glutathione S-transferase (GST) theta 1 (GSTT1), is a member of a superfamily of proteins that catalyze the conjugation of reduced glutathione to a variety of electrophilic and hydrophobic compounds. Human GSTs can be divided into five main classes: alpha, mu, pi, theta, and zeta. The theta class includes GSTT1 and GSTT2. GSTT1 and GSTT2 share 55% amino acid sequence identity and both may play an important role in human carcinogenesis. The GSTT1 and GSTT2 genes have a similar structure, being composed of five exons with identical exon/intron boundaries. This GSTT1 gene is haplotype-specific and is absent from 38% of the population. Alternative splicing of this gene results in multiple transcript variants. Two related pseudogenes, which are also located on chromosome 22, have been identified. [provided by RefSeq, Jun 2014]
