Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage, store at -20 degree C. Aliquots are stable for at least 6 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Urea is the major end product of nitrogen metabolism and it is concentrated up to four times higher than that of plasma. A rapid, passive transport of urea through the membranes of the IMCD is responsible for generation of high urea concentration in the inner medullary interstitium. Recently, a vasopressin-sensitive urea transporter (UT2; 397aa) from rabbit and its homolog from rat (rUT2; 397aa) have been cloned.

Uniprot Description

Function: Specialized low-affinity vasopressin-regulated urea transporter. Mediates rapid transepithelial urea transport across the inner medullary collecting duct and plays a major role in the urinary concentrating mechanism. Vasopressin regulates urea transport by increasing isoform 1 accumulation in the plasma membrane and/or phosphorylation of isoform 1. Ref.2 Ref.3 Ref.4 Ref.5

Subunit structure: Isoform 1 interacts with SNAPIN which may be important for recruitment to the plasma membrane. Ref.7

Subcellular location: Apical cell membrane; Multi-pass membrane protein Ref.7.

Tissue specificity: Isoform 1 and isoform 2 are expressed in the inner medulla of the kidney. Isoform 3 and isoform 4 are expressed in both the inner and outer renal medulla. Ref.1 Ref.2 Ref.3 Ref.4 Ref.5

Sequence similarities: Belongs to the urea transporter family.

Caution: It is uncertain whether Met-1 or Met-9 is the initiator.
