Rat Transcription factor HES-1 (HES1) ELISA Kit
Principle of the Assay||HES1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-HES1 antibody and an HES1 -HRP conjugate. The assay sample and buffer are incubated together with HES1 -HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the HES1 concentration since HES1 from samples and HES1 -HRP conjugate compete for the anti-HES1 antibody binding site. Since the number of sites is limited, as more sites are occupied by HES1 from the sample, fewer sites are left to bind HES1 -HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The HES1 concentration in each sample is interpolated from this standard curve.
NCBI and Uniprot Product Information
NCBI Description
This protein belongs to the basic helix-loop-helix family of transcription factors. It is a transcriptional repressor of genes that require a bHLH protein for their transcription. The protein has a particular type of basic domain that contains a helix interrupting protein that binds to the N-box rather than the canonical E-box. [provided by RefSeq, Jul 2008]
Uniprot Description
HES1: Transcriptional repressor of genes that require a bHLH protein for their transcription. May act as a negative regulator of myogenesis by inhibiting the functions of MYOD1 and ASH1. Binds DNA on N-box motifs: 5'-CACNAG-3' with high affinity and on E-box motifs: 5'-CANNTG-3' with low affinity. May play a role in a functional FA core complex response to DNA cross-link damage, being required for the stability and nuclear localization of FA core complex proteins, as well as for FANCD2 monoubiquitination in response to DNA damage.

The Rat HES1 hes1 (Catalog #AAA7229266) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7229266 ELISA Kit recognizes Rat HES1. It is sometimes possible for the material contained within the vial of "Transcription factor HES-1 (HES1), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
