Rat Transcription factor 4 (TCF4) ELISA Kit
Principle of the Assay||TCF4 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-TCF4 antibody and an TCF4-HRP conjugate. The assay sample and buffer are incubated together with TCF4-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the TCF4 concentration since TCF4 from samples and TCF4-HRP conjugate compete for the anti-TCF4 antibody binding site. Since the number of sites is limited, as more sites are occupied by TCF4 from the sample, fewer sites are left to bind TCF4-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The TCF4 concentration in each sample is interpolated from this standard curve.

This gene encodes transcription factor 4, a basic helix-loop-helix transcription factor. The encoded protein recognizes an Ephrussi-box ('E-box') binding site ('CANNTG') - a motif first identified in immunoglobulin enhancers. This gene is broadly expressed, and may play an important role in nervous system development. Defects in this gene are a cause of Pitt-Hopkins syndrome. Multiple alternatively spliced transcript variants that encode different proteins have been described. [provided by RefSeq, Aug 2011]

Activates transcription by binding to the E box (5'-CANNTG-3'). Binds to the E-box present in the somatostatin receptor 2 initiator element (SSTR2-INR) to activate transcription. Preferentially binds to either 5'-ACANNTGT-3' or 5'-CCANNTGG-3'. Efficient DNA binding requires dimerization with another bHLH protein. Forms homo- or heterooligomers with myogenin. Interacts with HIVEP2. Interacts with NEUROD2. Expressed in adult heart, brain, placenta, skeletal muscle and to a lesser extent in the lung. In developing embryonic tissues, expression mostly occurs in the brain. 3 isoforms of the human protein are produced by alternative splicing.
