Principle of the Assay: The ELISA kit adopts double antibody sandwich method: The rat TNF-alpha McAb was coated on the enzyme label plate and the rat TNF in the sample and standard binds to monoclonal antibody and the free components are washed away. Biotinylated anti rat TNF antibody was added, and horseradish peroxidase labeled Avidin was futher added. Biotin binds specifically to avidin; Anti rat TNF-alphaAntibody and rat TNF bound to monoclonal antibody will combine to form immune complex, and the free components are washed away. The chromogenic substrate is added, if there is rat TNF in the reaction well, horseradish peroxidase will make the colorless chromogenic agent blue and turn yellow after adding the termination solution. OD value was measured at 450 nm. The concentration of TNF-alphadetected in rats is directly proportional to the OD450 value, and the rat TNF in the sample can be calculated by drawing the standard curve TNF-alpha concentration.

Rat TNF- alpha is composed of an intracellular domain of 35 amino acids, a transmembrane domain of 21 amino acid residues and an extracellular domain of 179 amino acids. The extracellular domain has 94% amino acid sequence identity with mice and 69-76% sequence identity with cattle, dogs, horses, cats, humans, pigs and rhesus monkeys. The 26 kDa type II transmembrane protein is assembled in cells to form a non covalent connection The binding of the complex can induce reverse signal transduction, promote lymphocyte co stimulation and weaken the response of monocytes. It regulates the development of lymphoid tissue by controlling apoptosis. It can also promote inflammatory response by inducing activated vascular endothelial cells and macrophages. TNF- alpha is a key cytokine in the development of a variety of inflammatory diseases. A large number of foreign reports suggest that TNF- alpha may be an important mediator in the pathogenesis of insulin resistance. It can interfere with the insulin action of surrounding tissues and cause insulin resistance.
