Background/Introduction: Tumor necrosis factor alpha (TNF)- alpha), also known as cachectin and TNFSF1A, it is a pleiotropic cytokine that plays an important role in inflammation, apoptosis and immune system development.It can be produced by a variety of immune cells and epithelial cells. Rat TNF- alpha is composed of an intracellular domain of 35 amino acids, a transmembrane domain of 21 amino acid residues and an extracellular domain of 179 amino acids. The extracellular domain has 94% amino acid sequence identity with mice and 69-76% sequence identity with cattle, dogs, horses, cats, humans, pigs and rhesus monkeys. The 26 kDa type II transmembrane protein is assembled in cells to form a non covalent connection The binding of the complex can induce reverse signal transduction, promote lymphocyte co stimulation and weaken the response of monocytes. It regulates the development of lymphoid tissue by controlling apoptosis. It can also promote inflammatory response by inducing activated vascular endothelial cells and macrophages. TNF- alpha is a key cytokine in the development of a variety of inflammatory diseases. A large number of foreign reports suggest that TNF- alpha may be an important mediator in the pathogenesis of insulin resistance. It can interfere with the insulin action of surrounding tissues and cause insulin resistance.
