Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Description: This Rat [Pyruvate dehydrogenase [lipoamide]] kinase isozyme 1, mitochondrial (PDK1) ELISA Kit employs a two-site sandwich ELISA to quantitate PDK1 in samples. An antibody specific for PDK1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any PDK1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PDK1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PDK1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: PDK1 codes for an isozyme of pyruvate dehydrogenase kinase (PDK).The human PDK1 gene encodes a protein with a predicted molecular mass of 49,244 Da that shares 93% identity with that of the rat PDK1 gene. Northern blot analysis showed that the PDK1 message was expressed predominantly in the heart. Pyruvate dehydrogenase (PDH) is a part of a mitochondrial multienzyme complex that catalyzes the oxidative decarboxylation of pyruvate and is one of the major enzymes responsible for the regulation of homeostasis of carbohydrate fuels in mammals. The enzymatic activity is regulated by a phosphorylation/dephosphorylation cycle. Phosphorylation of PDH by a specific pyruvate dehydrogenase kinase (PDK) results in inactivation.

Kinase that plays a key role in regulation of glucose and fatty acid metabolism and homeostasis via phosphorylation of the pyruvate dehydrogenase subunits PDHA1 and PDHA2. This inhibits pyruvate dehydrogenase activity, and thereby regulates metabolite flux through the tricarboxylic acid cycle, down-regulates aerobic respiration and inhibits the formation of acetyl-coenzyme A from pyruvate. Plays an important role in cellular responses to hypoxia and is important for cell proliferation under hypoxia. Protects cells against apoptosis in response to hypoxia and oxidative stress.
