Intended Uses: This ATP2B1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Rat ATP2B1. This ELISA kit for research use only!

Principle of the Assay: ATP2B1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ATP2B1 antibody and an ATP2B1-HRP conjugate. The assay sample and buffer are incubated together with ATP2B1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ATP2B1 concentration since ATP2B1 from samples and ATP2B1-HRP conjugate compete for the anti-ATP2B1 antibody binding site. Since the number of sites is limited, as more sites are occupied by ATP2B1 from the sample, fewer sites are left to bind ATP2B1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ATP2B1 concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene belongs to the family of P-type primary ion transport ATPases characterized by the formation of an aspartyl phosphate intermediate during the reaction cycle. These enzymes remove bivalent calcium ions from eukaryotic cells against very large concentration gradients and play a critical role in intracellular calcium homeostasis. The mammalian plasma membrane calcium ATPase isoforms are encoded by at least four separate genes and the diversity of these enzymes is further increased by alternative splicing of transcripts. The expression of different isoforms and splice variants is regulated in a developmental, tissue- and cell type-specific manner, suggesting that these pumps are functionally adapted to the physiological needs of particular cells and tissues. This gene encodes the plasma membrane calcium ATPase isoform 1. Alternatively spliced transcript variants encoding different isoforms have been identified. [provided by RefSeq, Jul 2008]
