Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Related Product Information for PTHLH elisa kit
Description: This Rat Parathyroid Hormone-Like Related Protein (PTHLH) ELISA Kit employs a two-site sandwich ELISA to quantitate PTHLH in samples. An antibody specific for PTHLH has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any PTHLH present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PTHLH is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PTHLH bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: Parathyroid hormone-related protein (or PTHrP) is a protein occasionally secreted by cancer cells (breast cancer, certain types of lung cancer including squamous cell carcinoma). However, it also has normal functions. PTHrP acts as an endocrine, autocrine/ paracrine, and intracrine hormone. PTHrP is related in function to the "normal" parathyroid hormone. When a tumor secretes PTHrP, this can lead to hypercalcemia. As this is sometimes the first sign of the malignancy, hypercalcemia caused by PTHrP is considered a paraneoplastic phenomenon. PTHrP shares the same N-terminal end as parathyroid hormone and therefore it can bind to the same receptor (the Type I PTH receptor). PTHrP aids in normal mammary gland development and lactation as well as placental transfer of calcium.

Uniprot Description

Neuroendocrine peptide which is a critical regulator of cellular and organ growth, development, migration, differentiation and survival and of epithelial calcium ion transport. Regulates endochondral bone development and epithelial-mesenchymal interactions during the formation of the mammary glands and teeth. Required for skeletal homeostasis. Promotes mammary mesenchyme differentiation and bud outgrowth by modulating mesenchymal cell responsiveness to BMPs. Upregulates BMPR1A expression in the mammary mesenchyme and this increases the sensitivity of these cells to BMPs and allows them to respond to BMP4 in a paracrine and/or autocrine fashion. BMP4 signaling in the mesenchyme, in turn, triggers epithelial outgrowth and augments MSX2 expression, which causes the mammary mesenchyme to inhibit hair follicle formation within the nipple sheath ().

Product Notes

The Rat PTHLH pthlh (Catalog #AAA9712022) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9712022 ELISA Kit recognizes Rat PTHLH. It is sometimes possible for the material contained within the vial of "Parathyroid Hormone-Like Related Protein (PTHLH), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
