Rat Myc proto-oncogene protein ELISA Kit
Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to Myc proto-oncogene protein. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated polyclonal antibody preparation specific for Myc proto-oncogene protein and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. Only those wells that contain Myc proto-oncogene protein, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of Myc proto-oncogene protein in the samples is then determined by comparing the O.D. of the samples to the standard curve.

The protein encoded by this gene is a multifunctional, nuclear phosphoprotein that plays a role in cell cycle progression, apoptosis and cellular transformation. It functions as a transcription factor that regulates transcription of specific target genes. Mutations, overexpression, rearrangement and translocation of this gene have been associated with a variety of hematopoietic tumors, leukemias and lymphomas, including Burkitt lymphoma, in human. There is evidence to show that alternative translation initiations from an upstream, in-frame non-AUG (CUG) and a downstream AUG start site result in the production of two isoforms with distinct N-termini, in human and mouse. Rat mRNA also has a similarly placed CUG upstream of the AUG start site, suggesting that it may also produce two Myc proteins. [provided by RefSeq, Jul 2008]
