Rat Interleukin-12 receptor subunit beta-2 (IL12RB2) ELISA Kit
Principle of the Assay: IL-12RB2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-IL-12RB2 antibody and an IL-12RB2-HRP conjugate. The assay sample and buffer are incubated together with IL-12RB2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the IL-12RB2 concentration since IL-12RB2 from samples and IL-12RB2-HRP conjugate compete for the anti-IL-12RB2 antibody binding site. Since the number of sites is limited, as more sites are occupied by IL-12RB2 from the sample, fewer sites are left to bind IL-12RB2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The IL-12RB2 concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is a type I transmembrane protein identified as a subunit of the interleukin 12 receptor complex. The coexpression of this and IL12RB1 proteins was shown to lead to the formation of high-affinity IL12 binding sites and reconstitution of IL12 dependent signaling. The expression of this gene is up-regulated by interferon gamma in Th1 cells, and plays a role in Th1 cell differentiation. The up-regulation of this gene is found to be associated with a number of infectious diseases, such as Crohn's disease and leprosy, which is thought to contribute to the inflammatory response and host defense.
