Principle of the Assay||CYP2B6 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-CYP2B6 antibody and an CYP2B6-HRP conjugate. The assay sample and buffer are incubated together with CYP2B6-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CYP2B6 concentration since CYP2B6 from samples and CYP2B6-HRP conjugate compete for the anti-CYP2B6 antibody binding site. Since the number of sites is limited, as more sites are occupied by CYP2B6 from the sample, fewer sites are left to bind CYP2B6-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CYP2B6 concentration in each sample is interpolated from this standard curve.

This gene, CYP2B6, encodes a member of the cytochrome P450 superfamily of enzymes. The cytochrome P450 proteins are monooxygenases which catalyze many reactions involved in drug metabolism and synthesis of cholesterol, steroids and other lipids. This protein localizes to the endoplasmic reticulum and its expression is induced by phenobarbital. The enzyme is known to metabolize some xenobiotics, such as the anti-cancer drugs cyclophosphamide and ifosphamide. Transcript variants for this gene have been described; however, it has not been resolved whether these transcripts are in fact produced by this gene or by a closely related pseudogene, CYP2B7. Both the gene and the pseudogene are located in the middle of a CYP2A pseudogene found in a large cluster of cytochrome P450 genes from the CYP2A, CYP2B and CYP2F subfamilies on chromosome 19q. It oxidizes a variety of structurally unrelated compounds, including steroids, fatty acids, and xenobiotics. Phosphorylation is accompanied by a decrease in enzyme activity.
