Principle of the Assay: CTHRC1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-CTHRC1 antibody and an CTHRC1-HRP conjugate. The assay sample and buffer are incubated together with CTHRC1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CTHRC1 concentration since CTHRC1 from samples and CTHRC1-HRP conjugate compete for the anti-CTHRC1 antibody binding site. Since the number of sites is limited, as more sites are occupied by CTHRC1 from the sample, fewer sites are left to bind CTHRC1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CTHRC1 concentration in each sample is interpolated from this standard curve.

CTHRC1: May act as a negative regulator of collagen matrix deposition. Defects in CTHRC1 may be a cause of Barrett esophagus (BE). A condition characterized by a metaplastic change in which normal esophageal squamous epithelium is replaced by a columnar and intestinal-type epithelium. Patients with Barrett esophagus have an increased risk of esophageal adenocarcinoma. The main cause of Barrett esophagus is gastroesophageal reflux. The retrograde movement of acid and bile salts from the stomach into the esophagus causes prolonged injury to the esophageal epithelium and induces chronic esophagitis, which in turn is believed to trigger the pathologic changes. Genetic variants in CTHRC1 have been found in individuals with Barrett esophagus and are thought to contribute to disease susceptibility. 2 isoforms of the human protein are produced by alternative splicing.
