Principle of the assay: CLPS ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CLPS antibody and an CLPS-HRP conjugate. The assay sample and buffer are incubated together with CLPS-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CLPS concentration since CLPS from samples and CLPS-HRP conjugate compete for the anti-CLPS antibody binding site. Since the number of sites is limited, as more sites are occupied by CLPS from the sample, fewer sites are left to bind CLPS-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CLPS concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is a cofactor needed by pancreatic lipase for efficient dietary lipid hydrolysis. It binds to the C-terminal, non-catalytic domain of lipase, thereby stabilizing an active conformation and considerably increasing the overall hydrophobic binding site. The gene product allows lipase to anchor noncovalently to the surface of lipid micelles, counteracting the destabilizing influence of intestinal bile salts. This cofactor is only expressed in pancreatic acinar cells, suggesting regulation of expression by tissue-specific elements. Three transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Nov 2011]
