Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Related Product Information for TNFAIP1 elisa kit
Description: This Rat BTB/POZ domain-containing adapter for CUL3-mediated RhoA degradation protein 2 (TNFAIP1) ELISA Kit employs a two-site sandwich ELISA to quantitate TNFAIP1 in samples. An antibody specific for TNFAIP1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any TNFAIP1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TNFAIP1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TNFAIP1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: TNFAIP1 was identified as a gene whose expression can be induced by the tumor necrosis factor alpha (TNF) in umbilical vein endothelial cells. Studies of a similar gene in mouse suggest that the expression of this gene is developmentally regulated in a tissue-specific manner. Wolf et al. (1992) characterized a novel cDNA by differential screening of a tumor necrosis factor-alpha (TNFA)-stimulated umbilical vein endothelial cell library. The protein product is involved in the primary response of the endothelium to TNF. The 3.5-kb transcript was found to be expressed in a developmentally regulated, tissue-specific manner. The gene, present in single copy, was located in the 17q22-q23 region by in situ hybridization.

Uniprot Description

TNFAIP1: Substrate-specific adapter of a BCR (BTB-CUL3-RBX1) E3 ubiquitin-protein ligase complex involved in regulation of cytoskeleton structure. The BCR(BACURD2) E3 ubiquitin ligase complex mediates the ubiquitination of RHOA, leading to its degradation by the proteasome, thereby regulating the actin cytoskeleton and cell migration. Its interaction with RHOB may regulate apoptosis. May enhance the PCNA-dependent DNA polymerase delta activity. Belongs to the BACURD family.

Protein type: DNA replication; Ubiquitin conjugating system

Cellular Component: cytoplasm; nucleolus; endosome

Molecular Function: protein domain specific binding; protein binding; ubiquitin-protein ligase activity; GTP-Rho binding

Biological Process: proteasomal ubiquitin-dependent protein catabolic process; cell migration; protein heterooligomerization; protein ubiquitination; DNA repair; negative regulation of Rho protein signal transduction; regulation of transcription, DNA-dependent; stress fiber formation; immune response; actin cytoskeleton organization and biogenesis; DNA replication; positive regulation of DNA replication; protein homooligomerization

Product Notes

The Rat TNFAIP1 tnfaip1 (Catalog #AAA9711677) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9711677 ELISA Kit recognizes Rat TNFAIP1. It is sometimes possible for the material contained within the vial of "BTB/POZ Domain-Containing Adapter For CUL3-Mediated RhoA Degradation Protein 2 (TNFAIP1), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
