Principle of the Assay: Rat Amyloid beta (Abeta) ELISA Kit employs a two-site sandwich ELISA to quantitate Abeta in samples. An antibody specific for Abeta has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any Abeta present is bound by the immobilized antibody. After removing any unbound substances, HRP-Conjugated Rat Abeta detection antibody is added to the wells. Following a wash to remove any unbound HRP reagent, a Chromogen solution is added to the wells and color develops in proportion to the amount of Abeta bound in the initial step. The color development is stopped and the intensity of the color is measured.
Background/Introduction: Amyloid beta (Abeta or Abeta) denotes peptides of 36-43 amino acids that are crucially involved in Alzheimer's disease as the main component of the amyloid plaques found in the brains of Alzheimer patients. The peptides derive from the amyloid precursor protein (APP), which is cleaved by beta secretase and gamma secretase to yield Abeta. Abeta molecules can aggregate to form flexible soluble oligomers which may exist in several forms.
