Principle of the Assay: ADAMTS3 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-ADAMTS3 antibody and an ADAMTS3-HRP conjugate. The assay sample and buffer are incubated together with ADAMTS3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ADAMTS3 concentration since ADAMTS3 from samples and ADAMTS3-HRP conjugate compete for the anti-ADAMTS3 antibody binding site. Since the number of sites is limited, as more sites are occupied by ADAMTS3 from the sample, fewer sites are left to bind ADAMTS3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ADAMTS3 concentration in each sample is interpolated from this standard curve.

This gene encodes a member of the ADAMTS (a disintegrin and metalloproteinase with thrombospondin motifs) protein family. Members of the family share several distinct protein modules, including a propeptide region, a metalloproteinase domain, a disintegrin-like domain, and a thrombospondin type 1 (TS) motif. Individual members of this family differ in the number of C-terminal TS motifs, and some have unique C-terminal domains. The protein encoded by this gene is the major procollagen II N-propeptidase. A deficiency of this protein may be responsible for dermatosparaxis, a genetic defect of connective tissues. [provided by RefSeq, Jul 2008]
