Principle of the Assay: Rac1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-Rac1 antibody and an Rac1-HRP conjugate. The assay sample and buffer are incubated together with Rac1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the Rac1 concentration since Rac1 from samples and Rac1-HRP conjugate compete for the anti-Rac1 antibody binding site. Since the number of sites is limited, as more sites are occupied by Rac1 from the sample, fewer sites are left to bind Rac1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The Rac1 concentration in each sample is interpolated from this standard curve.

Uniprot Description

RAC1: a plasma membrane-associated member of the Rho-GTPase family. Plays a key role in cytoskeletal reorganization, membrane trafficking, transcriptional regulation and cell growth and development. GTP binding stimulates its activity. Phosphorylation by Akt may inhibit GTP binding of Rac1, therefore attenuating the downstream signal transduction pathway. Found in a trimeric complex composed of DOCK1 and ELMO1, which plays a central role in phagocytosis of apoptotic cells. Two alternatively spliced isoforms have been described.

Product Notes

The Monkey RAC1 rac1 (Catalog #AAA750101) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA750101 ELISA Kit recognizes Monkey RAC1. It is sometimes possible for the material contained within the vial of "Ras-related C3 Botulinum Toxin Substrate 1, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
