Intended Uses: This PS1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Rat PS1. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: PS1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-PS1 antibody and an PS1-HRP conjugate. The assay sample and buffer are incubated together with PS1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PS1 concentration since PS1 from samples and PS1-HRP conjugate compete for the anti-PS1 antibody binding site. Since the number of sites is limited, as more sites are occupied by PS1 from the sample, fewer sites are left to bind PS1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PS1 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a trypsinogen, which is a member of the trypsin family of serine proteases. This enzyme is secreted by the pancreas and cleaved to its active form in the small intestine. It is active on peptide linkages involving the carboxyl group of lysine or arginine. Mutations in this gene are associated with hereditary pancreatitis. This gene and several other trypsinogen genes are localized to the T cell receptor beta locus on chromosome 7. [provided by RefSeq, Jul 2008]

A disease characterized by the presence of calculi in pancreatic ducts. It causes severe abdominal pain attacks. Belongs to the peptidase S1 family.
