Specificity
A 21 amino acid control peptide for MBS618407 near the N-terminus of mouse Agouti (1). The sequence shows no significant sequence homology with AGRP, tubby, tub, TULP1 or TULP2. Species Sequence Homology: rat; 90%, human and monkey; 80%, porcine; 73%, fox; 75%, bovine; 66%

Suitable for use in ELISA and antibody blocking experiments. Not recommended for use in Western Blot. Other applications not tested. Optimal dilutions to be determined by the researcher.
Preparation and Storage
Lyophilized and reconstituted products are stable for 6 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Agouo protein (132 aa in human) is normally expressed in skin but its ubiquitous expression causes obesity. Agouti is a paracrine-signaling molecule that affects pigmentation by inhibiting the melonocortin receptor 1 (MVC-1). However, recombinant agouti protien also antagonizes the MC2R and MC4R. ARGP (132aa in human, chromosomes 16q21) is normally expressed in adrenal and hypothalamus. AGRP levels are increased several folds in ob/ob mice. AGRP is a strong antagonist of MC3R ans MV4R. Ubiquitous expression of AGRP in transgenic mice causes obesity without altering skin pigmentation.

NCBI Description

In mice, the agouti gene encodes a paracrine signaling molecule that causes hair follicle melanocytes to synthesize pheomelanin, a yellow pigment, instead of the black or brown pigment, eumelanin. Pleiotropic effects of constitutive expression of the mouse gene include adult-onset obesity, increased tumor susceptibility, and premature infertility. This gene is highly similar to the mouse gene and encodes a secreted protein that may (1) affect the quality of hair pigmentation, (2) act as a pharmacological antagonist of alpha-melanocyte-stimulating hormone, (3) play a role in neuroendocrine aspects of melanocortin action, and (4) have a functional role in regulating lipid metabolism in adipocytes. ASIP: Involved in the regulation of melanogenesis. The binding of ASP to MC1R precludes alpha-MSH initiated signaling and thus blocks production of cAMP, leading to a down-regulation of eumelanogenesis (brown/black pigment) and thus increasing synthesis of pheomelanin (yellow/red pigment). In higher primates, agouti may affect the quality of hair pigmentation rather than its pattern of deposition. Could well play a role in neuroendocrine aspects of melanocortin action. May have some functional role in regulating the lipid metabolism with adipocytes.
