Related Product Information for PRLR recombinant protein
Background: Prolactin receptor (PRLR) is a single-pass transmembrane receptor belonging to the type I  cytokine receptor superfamily, and contains two fibronectin type -I¢ domains. All class 1 ligands activate their respective receptors by clustering mechanisms. Ligand binding results in the transmembrane PRLR dimerization, followed by phosphorylation and activation of the molecules invloved in the signaling pathways, such as Jak-STAT, Ras/Raf/MAPK. The PRLR contains no intrinsic tyrosine kinase cytoplasmic domain but associates with a cytoplasmic tyrosine kinase, JAK2. PRLR mainly serves as the receptor for the pituitary hormone prolactin (PRL), a secreted hormone that affects reproduction and homeostasis in vertebrates. PRLR can be regulated by an interplay of two different mechanisms, PRL or ovarian steroid hormones independently or in combination in a tissue-specific manner. The role of the hormone prolactin (PRL) in the pathogenesis of breast cancer is mediated by its cognate receptor (PRLR). Ubiquitin-dependent degradation of the PRLR that negatively regulates PRL signaling is triggered by PRL-mediated phosphorylation of PRLR on Ser349 followed by the recruitment of the beta-transducin repeats-containing protein (beta-TrCP) ubiquitin-protein isopeptide ligase. which altered PRLR stability may directly influence the pathogenesis of breast cancer.

Description: A DNA sequence encoding the extracellular domain of mouse PRLR (NP_035299.4) (Met 1-Asp 229) was fused with the C-terminal polyhistidine-tagged Fc region of human IgG1 at the C-terminus.

Uniprot Description

PRLR: receptor for the anterior pituitary hormone prolactin and placental lactogen I and II. Interacts with Cyclophilin A, differentially regulating various signaling pathways from the PrlR. Prolactin signaling is attenuated by threonine and tyrosine phosphorylation. Two alternative splice isoforms have been identified.

Product Notes

The PRLR prlr (Catalog #AAA2546975) is a Recombinant Protein produced from Human Cells and is intended for research purposes only. The product is available for immediate purchase. The recombinant mouse PRLR/Fc chimera is a disulfide-linked homodimer. The reduced monomer consists of 457 amino acids and has a calculated molecular mass of 52.5 kDa. As a result of glycosylation, the apparent molecular mass of the mouse PRLR/Fc monomer is approximately 65-70 kDa in SDS-PAGE under reducing conditions. Researchers should empirically determine the suitability of the PRLR prlr for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "PRLR / Prolactin Receptor, Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
