Description: Recombinant full length human PP2Aalpha and human PPP2R1A were expressed by baculovirus in Sf9 insect cells using an N-terminal His tag.
Scientific Background: PP2Aalpha is also known as Protein phosphatase 2A active subunit. PP2A is one of the four major Ser/Thr phosphatases and is a human tumor suppressor that inhibits cellular transformation. PP2A alpha gene encodes a specific phosphotyrosyl phosphatase activator of the dimeric form of protein phosphatase-2A (1). Protein phosphatase 2A holoenzyme is a heterotrimeric proteins composed of a structural subunit A, a catalytic subunit C, and a regulatory subunit B. PTP2A alpha can alter the substrate specificity of PP2A leading to enhanced phosphotyrosine phosphatase activity and decreased phosphoserine phosphatase activity (2).

NCBI Description

This gene encodes the phosphatase 2A catalytic subunit. Protein phosphatase 2A is one of the four major Ser/Thr phosphatases, and it is implicated in the negative control of cell growth and division. It consists of a common heteromeric core enzyme, which is composed of a catalytic subunit and a constant regulatory subunit, that associates with a variety of regulatory subunits. PPP2CA: ubiquitous cytosolic Ser/Thr protein phosphatase, catalytic subunit. The holoenzyme consists of a 36 kDa catalytic subunit (subunit C) and a 65 kDa constant regulatory subunit (PR65 or subunit A), that associates with a variety of regulatory subunits. Proteins that associate with the core dimer include three families of regulatory subunits B (the R2/B/PR55/B55, R3/B'/PR72/PR130/PR59 and R5/B'/B56 families), the 48 kDa variable regulatory subunit, viral proteins, and cell signaling molecules.
