PPCDC belongs to the HFCD (homo oligomeric flavin containing Cys decarboxylase) superfamily that plays a role in the biosynthesis of coenzyme A (CoA) from pantothenate (Vitamin B). The biosynthesis of CoA from pantothenate requires several steps: the phosphorylation of pantothenate, the conversion of 4'-hosphopantothenate to 4'-phosphopantetheine, the adenylation by phosphopantetheine adenylyltransferase to form dephospho-CoA and the phosphorylation by dephospho-CoA kinase to form CoA. PPCDC plays a direct role in this pathway by converting 4'-phosphopantothenate into 4'-phosphopantetheine. Recombinant human PPCDC protein, fused to His-tag at N-terminus, was expressed in E Coli and purified by using conventional chromatography techniques.

Biosynthesis of coenzyme A (CoA) from pantothenic acid (vitamin B5) is an essential universal pathway in prokaryotes and eukaryotes. PPCDC (EC 4.1.1.36), one of the last enzymes in this pathway, converts phosphopantothenoylcysteine to 4-prime-phosphopantetheine (Daugherty et al., 2002 [PubMed 11923312]).[supplied by OMIM, Mar 2008]
