Principle of the Assay: The MyBioSource' Human Periostin ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human Periostin in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Human Periostin coated on a 96-well plate. Standards and samples are pipetted into the wells and Periostin present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human Periostin antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Periostin bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background/Introduction: Periostin, also known as OSF2, is a protein that in humans is encoded by the POSTN gene. The International Radiation Hybrid Mapping Consortium mapped the POSTN gene to chromosome 13. Periostin functions as a ligand for alpha-V/beta-3 and alpha-V/beta-5 integrins to support adhesion and migration of epithelial cells. It is found that periostin was overexpressed by the majority of human primary breast cancers examined. After myocardial infarction, periostin-induced cardiomyocyte cell cycle reentry and mitosis were associated with improved ventricular remodeling and myocardial function, reduced fibrosis and infarct size, and increase angiogenesis.

NCBI Description

This gene encodes a secreted extracellular matrix protein that functions in tissue development and regeneration, including wound healing, and ventricular remodeling following myocardial infarction. The encoded protein binds to integrins to support adhesion and migration of epithelial cells. This protein plays a role in cancer stem cell maintenance and metastasis. Mice lacking this gene exhibit cardiac valve disease, and skeletal and dental defects. Alternative splicing results in multiple transcript variants encoding different isoforms. [provided by RefSeq, Sep 2015]

Uniprot Description

POSTN: Binds to heparin. Induces cell attachment and spreading and plays a role in cell adhesion. May play a role in extracellular matrix mineralization. 4 isoforms of the human protein are produced by alternative splicing.

Protein type: Secreted; Cell adhesion; Secreted, signal peptide; Motility/polarity/chemotaxis

Chromosomal Location of Human Ortholog: 13q13.3

Cellular Component: extracellular matrix; proteinaceous extracellular matrix; extracellular space; trans-Golgi network

Molecular Function: heparin binding; protein binding; cell adhesion molecule binding

Biological Process: tissue development; extracellular matrix organization and biogenesis; regulation of Notch signaling pathway; cell adhesion; skeletal development

Product Notes

The Human POSTN postn (Catalog #AAA824679) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA824679 ELISA Kit recognizes Human POSTN. AAA Biotech's Periostin can be used in a range of immunoassay formats including, but not limited to, Sandwich ELISA (SE). Researchers should empirically determine the suitability of the POSTN postn for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Periostin, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
