Description: This assay employs a two-site sandwich ELISA to quantitate MC2R in samples. An antibody specific for MC2R has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MC2R present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MC2R is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MC2R bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: The ACTH receptor is a type of melanocortin receptor (type 2), properly known as MC2, which is specific for ACTH.the ACTHR gene is expressed in normal corporal skin, scalp tissue, compound nevus tissue, basal cell carcinoma tissue, and cultured melanocytes; the CYP11A1 gene is expressed in squamous cell carcinoma tissue, normal facial skin, normal corporal skin, cultured melanocytes, and basal cell carcinoma tissue; the CYP17 gene is expressed in normal facial skin, corporal skin, cultured melanocytes, squamous cell carcinoma cells, and basal cell carcinoma cells; the CYP21A2 gene is expressed in melanoma tissue, squamous cell carcinoma tissue, basal cell carcinoma tissue, normal facial skin, and cultured melanocytes.
