Intended Uses: This MC2R ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse MC2R. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: MC2R ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-MC2R antibody and an MC2R-HRP conjugate. The assay sample and buffer are incubated together with MC2R-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the MC2R concentration since MC2R from samples and MC2R-HRP conjugate compete for the anti-MC2R antibody binding site. Since the number of sites is limited, as more sites are occupied by MC2R from the sample, fewer sites are left to bind MC2R-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The MC2R concentration in each sample is interpolated from this standard curve.

NCBI Description

MC2R encodes one member of the five-member G-protein associated melanocortin receptor family. Melanocortins (melanocyte-stimulating hormones and adrenocorticotropic hormone) are peptides derived from pro-opiomelanocortin (POMC). MC2R is selectively activated by adrenocorticotropic hormone, whereas the other four melanocortin receptors recognize a variety of melanocortin ligands. Mutations in MC2R can result in familial glucocorticoid deficiency. Alternate transcript variants have been found for this gene. [provided by RefSeq, May 2014]

MC2R: Receptor for ACTH. This receptor is mediated by G proteins (G(s)) which activate adenylate cyclase. Defects in MC2R are the cause of glucocorticoid deficiency type 1 (GCCD1); also known as familial glucocorticoid deficiency type 1 (FGD1). GCCD1 is an autosomal recessive disorder due to congenital insensitivity or resistance to adrenocorticotropin (ACTH). It is characterized by progressive primary adrenal insufficiency, without mineralocorticoid deficiency. Belongs to the G-protein coupled receptor 1 family.
