Principle of the Assay: GSTM3 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-GSTM3 antibody and an GSTM3-HRP conjugate. The assay sample and buffer are incubated together with GSTM3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GSTM3 concentration since GSTM3 from samples and GSTM3-HRP conjugate compete for the anti-GSTM3 antibody binding site. Since the number of sites is limited, as more sites are occupied by GSTM3 from the sample, fewer sites are left to bind GSTM3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GSTM3 concentration in each sample is interpolated from this standard curve.

Cytosolic and membrane-bound forms of glutathione S-transferase are encoded by two distinct supergene families. At present, eight distinct classes of the soluble cytoplasmic mammalian glutathione S-transferases have been identified: alpha, kappa, mu, omega, pi, sigma, theta and zeta. This gene encodes a glutathione S-transferase that belongs to the mu class. The mu class of enzymes functions in the detoxification of electrophilic compounds, including carcinogens, therapeutic drugs, environmental toxins and products of oxidative stress, by conjugation with glutathione. The genes encoding the mu class of enzymes are organized in a gene cluster on chromosome 1p13.3 and are known to be highly polymorphic. These genetic variations can change an individual's susceptibility to carcinogens and toxins as well as affect the toxicity and efficacy of certain drugs. Mutations of this class mu gene have been linked with a slight increase in a number of cancers, likely due to exposure with environmental toxins.
