Intended Uses: This YME1L1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Porcine YME1L1. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: YME1L1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-YME1L1 antibody and an YME1L1-HRP conjugate. The assay sample and buffer are incubated together with YME1L1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the YME1L1 concentration since YME1L1 from samples and YME1L1-HRP conjugate compete for the anti-YME1L1 antibody binding site. Since the number of sites is limited, as more sites are occupied by YME1L1 from the sample, fewer sites are left to bind YME1L1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The YME1L1 concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is the human ortholog of yeast mitochondrial AAA metalloprotease, Yme1p. It is localized in the mitochondria and can functionally complement a yme1 disruptant yeast strain. It is proposed that this gene plays a role in mitochondrial protein metabolism and could be involved in mitochondrial pathologies. Three transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Dec 2011]

Ensures cell proliferation, maintains normal cristae morphology and complex I respiration activity, promotes antiapoptotic activity and protects mitochondria from the accumulation of oxidatively damaged membrane proteins. Requires to control the accumulation of nonassembled respiratory chain subunits (NDUFB6, OX4 and ND1). Seems to act in the processing of OPA1. 2 isoforms of the human protein are produced by alternative splicing.
