Application Notes
Suitable for use in ELISA and Western Blot.
Dilution: Western Blot: 1-10ug/ml using ECL. Detects a major band at ~55 kD in postnuclear supernatants of CHO transfected with VMAT1 and not in wild type cells (2). Some additional bands, both higher and lower MW, were also detected and remain unaffected by the inclusion of protease inhibitors.
ELISA: 0.5-1ug/ml. Control peptide can be used to coat ELISA plates at 1ug/ml.
Immunogen
A 20aa peptide sequence near the C-terminal, cytoplasmic region of human VMAT1.
Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage, store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

NCBI Description

The vesicular monoamine transporter acts to accumulate cytosolic monoamines into synaptic vesicles, using the proton gradient maintained across the synaptic vesicular membrane. Its proper function is essential to the correct activity of the monoaminergic systems that have been implicated in several human neuropsychiatric disorders. The transporter is a site of action of important drugs, including reserpine and tetrabenazine (summary by Peter et al., 1993 [PubMed 7905859]).
