Preparation and Storage
Store product at 4 degree C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20 degree C. Aliquots are stable at -20 degree C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Immunofluorescence (IF)

(Fluorescent confocal image of Hela cell stained with TRIM28 Antibody (N-term).Hela cells were fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.1%, 10 min), then incubated with TRIM28 primary antibody (1:25, 1 h at 37 degree C). For secondary antibody, Alexa Fluor 488 conjugated donkey anti-rabbit antibody (green) was used (1:400, 50 min at 37 degree C).Cytoplasmic actin was counterstained with Alexa Fluor 555 (red) conjugated Phalloidin (7units/ml, 1 h at 37 degree C). Nuclei were counterstained with DAPI (blue) (10 ug/ml, 10 min). TRIM28 immunoreactivity is localized to Nucleus significantly. )

The protein encoded by this gene mediates transcriptional control by interaction with the Kruppel-associated box repression domain found in many transcription factors. The protein localizes to the nucleus and is thought to associate with specific chromatin regions. The protein is a member of the tripartite motif family. Mediates transcriptional regulation by interaction with the Kruppel-associated box (KRAB) repression domain found in many transcription factors. Binds to the negative regulatory domain of c-Myb and negatively regulates the c-Myb-dependent trans-activation. Localizes to the nucleus and is thought to associate with specific chromatin regions. Phosphorylated in an ATM-dependent manner following double-strand breaks. Its phosphorylation in response to DNA damage leads to chromatin relaxation. A member of the tripartite motif family. The tripartite motif includes three zinc-binding domains, a RING, a B-box type 1 and a B-box type 2, and a coiled-coil region.
