NCBI Description

Uniprot Description

TOPBP1: a DNA-binding protein and scaffolding protein that is required for DNA replication. Plays a role in the rescue of stalled replication forks and checkpoint control. Binds double-stranded DNA breaks and nicks as well as single-stranded DNA. Recruits the SWI/SNF chromatin remodeling complex to E2F1-responsive promoters. Down-regulates E2F1 activity and inhibits E2F1-dependent apoptosis during G1/S transition and after DNA damage. Induces a large increase in the kinase activity of ATR. Contains 7 BRCT domains, which are characteristic of proteins involved in cell cycle checkpoint control in response to DNA damage. Colocalizes with ATR and H2AFX at unsynapsed chromosome cores during prophase. Has a uniform nuclear distribution during G phase. Colocalizes with BRCA1 at stalled replication forks during S phase. In mitotic cells it colocalizes with BRCA1 at spindle poles and centrosomes during metaphase and anaphase. Detected in discrete foci together with PML and numerous DNA repair enzymes after DNA damage by alkylating agents, UV or gamma irradiation. Up-regulated during the S phase of the cell cycle. Up-regulated by E2F1 and interferon.

Product Notes

The TOPBP1 topbp1 (Catalog #AAA9411774) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The TOPBP1 Antibody reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's TOPBP1 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunohistochemistry (IHC), Immunofluorescence (IF). Western blotting: 1:500 - 1:2000 Immunohistochemistry: 1:50 - 1:200 Immunofluorescence: 1:50 - 1:200. Researchers should empirically determine the suitability of the TOPBP1 topbp1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "TOPBP1, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
