(Figure. Western blot analysis of TNF alpha using anti- TNF alpha antibody (MBS175453).Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V(Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.Lane: Recombinant Mouse TNFalpha Protein 0.5ng,After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti- TNF alpha antigen affinity purified polyclonal antibody at 0.5ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for TNF alpha at approximately 17KD. The expected band size for TNF alpha is at 17KD.)
Background: Tumor necrosis factor-alpha (TNFA) also known as TNFR1. It is a potent cytokine, elicits a broad spectrum of biological responses which are mediated by binding to a cell surface receptor. There are 2 different proteins that serve as major receptors for TNF-alpha, one associated with myeloid cells and one associated with epithelial cells. TNFR1 maps to 12p13. TNFR1 signaling is also known to activate the transcription factor NF-kappa B and promote survival.

This gene encodes a multifunctional proinflammatory cytokine that belongs to the tumor necrosis factor (TNF) superfamily. Members of this family are classified based on primary sequence, function, and structure. This protein is synthesized as a type-II transmembrane protein and is reported to be cleaved into products that exert distinct biological functions. It plays an important role in the innate immune response as well as regulating homeostasis but is also implicated in diseases of chronic inflammation. In mouse deficiency of this gene is associated with defects in response to bacterial infection, with defects in forming organized follicular dendritic cell networks and germinal centers, and with a lack of primary B cell follicles.
