Application Notes
ELISA: This purified human TIMP-1 antibody may be used in an indirect ELISA or as the capture reagent in a sandwich ELISA with our biotinylated human TIMP-1 antibody as the detection reagent and recombinant human TIMP-1 as the standard.
ELISA: Minimum Dilution: 0.5; Maximum Dilution: 2.0ug/ml
Immunohistology - Paraffin: Application Note: This product requires enzyme mediated antigen retrieval prior to staining of parrafin sections. Proteinase K is recomended for this purpose.
Western Blotting: Minimum Dilution: 0.1; Maximum Dilution: 0.2ug/ml
Reconstitution
Reconstitute with 0.1ml distilled water Care should be taken during reconstitution as the protein may appear as a film at the bottom of the vial. MyBioSource recommends that the vial is gently mixed after reconstitution. For long term storage the addition of 0.09% sodium azide is recommended.

Antiserum to human TIMP-1 were raised by repeated immunisation of rabbits with highly purified antigen. Purified IgG was prepared from whole serum by affinity chromatography.

Preparation and Storage
Prior to reconstitution store at 4 degree C. After reconstitution store at -20 degree C. Storage in frost-free freezers is not recommended. This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
Shelf Life: 12 months from date of reconstitution.

Rabbit anti Human TIMP-1 antibody specifically recognizes human TIMP-1 (Tissue Inhibitor of Metalloproteinase -1), a secreted glycoprotein and member of the protease inhibitor I35 (TIMP) family, which is expressed by a wide range of cells, including microvascular endothelial cells, smooth muscle cells, mesangial cells, and some fibroblasts. TIMP-1 acts as a natural extracellular inhibitor of MMPs (matrix metalloproteinases), a group of peptidases involved in degradation of the extracellular matrix, and is also involved in erythropoiesis, through the simulation of the proliferation/differentiation of erythroid progenitors. Studies have shown that a correlation between localized concentrations of TIMP-1 and TIMP-3, may determine cell fate, and that increased expression of TIMP-1, may act as an inhibitor/suppressor of invasive cell growth.

NCBI Description

This gene belongs to the TIMP gene family. The proteins encoded by this gene family are natural inhibitors of the matrix metalloproteinases (MMPs), a group of peptidases involved in degradation of the extracellular matrix. In addition to its inhibitory role against most of the known MMPs, the encoded protein is able to promote cell proliferation in a wide range of cell types, and may also have an anti-apoptotic function. Transcription of this gene is highly inducible in response to many cytokines and hormones. In addition, the expression from some but not all inactive X chromosomes suggests that this gene inactivation is polymorphic in human females. This gene is located within intron 6 of the synapsin I gene and is transcribed in the opposite direction. [provided by RefSeq, Jul 2008]

TIMP1: Complexes with metalloproteinases (such as collagenases) and irreversibly inactivates them by binding to their catalytic zinc cofactor. Also mediates erythropoiesis in vitro; but, unlike IL-3, it is species-specific, stimulating the growth and differentiation of only human and murine erythroid progenitors. Known to act on MMP-1, MMP-2, MMP-3, MMP-7, MMP-8, MMP-9, MMP-10, MMP-11, MMP-12, MMP-13 and MMP-16. Does not act on MMP-14. Belongs to the protease inhibitor I35 (TIMP) family.
