This antibody is directed against the C fragment of Tetanus toxin derived from Clostridium tetani. The product was prepared from monospecific antiserum by delipidation and defibrination. The antisera was tested by immunoblot analysis using recombinant TTFC with and without the polyhistidine tag (Roche Biosciences), and was found to be equally immunoreactive with both constructs.

Application Notes
This antibody has been tested for use in ELISA and western blot. Specific conditions for reactivity should be optimized by the end user. For western blot experiments, one can expect a band of 52 kDa in size corresponding to full-length TTFC. For direct ELISA, coat plates with 1 ug of recombinant TTFC (0.1ml/well). Antigen-specific signal can be detected at a dilution up to 1:10,000,000. For antigen capture ELISA, 0.1 ml of the diluted antisera (1:4000) was able to detect 6 picograms of TTFC using monoclonal mouse anti-TTFC as the antigen-detection antibody (0.5 ug/ml, Roche Biosciences). Sera diluted up to 100,000-fold can easily detect 600 picograms of recombinant TTFC by Western blot processed with ECL Plus Western Blotting and Detection System (Amersham Biosciences, 1 sec film exposure).
Immunogen
This antibody was prepared from whole rabbit serum produced by repeated immunizations with a polyhistidine-tagged TTFC fusion protein expressed in E Coli corresponding to full length TTFC. The polyhistidine-tag was placed at the amino terminus of the protein. The recombinant protein was purified from soluble bacterial extracts by metal-affinity and ion-exchange chromatography.
Preparation and Storage
This product is stable for several weeks at 4 degree C as an undiluted liquid. Dilute only prior to immediate use.
For extended storage, aliquot contents and freeze at-20 degree C or below. Avoid cycles of freezing and thawing. Expiration date is one (1) year from date of receipt.

The illness known as Tetanus is caused by a neurotoxin produced by the anaerobic bacterium Clostridium tetani. C. tetani is commonly found in soil, feces, and on many objects lying on the ground, such as rusty metal, hence puncture wounds are a common cause of tetanus. It acts upon the presynaptic membranes of both central and peripheral nervous systems to block the release of neurotransmitters. In its toxic form tetanus neurotoxin is a 150 kDa protein, consisting of two major components: a light chain and heavy chain. The light chain contains the enzymatic portion of the toxin and is responsible for its toxic effects. The heavy chain binds to the neuron and aids delivery of the light chain to the neuron interior. A single disulfide bond links light and heavy chains. Fragment C (TTFC) is that portion of the heavy chain that actually binds to gangliosides of the cell membrane. As such, TTFC is a non-toxic binding domain that can be used for cell targeting to increase the delivery of attached "passenger proteins" to neurons, both in vitro and in vivo. This antibody also has utility for most immunological experiments.

Researchers should empirically determine the suitability of the Tetanus Toxin Fragment C for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process.
